What is Phase Contrast Microscope?

A Phase Contrast microscope is a microscope used to observe unstained specimens. Living cells and unstained biological specimens, due to the difference in the refractive index and thickness of the fine structures of the cells, when the light wave passes through, the wavelength and amplitude do not change, only the phase changes (x phase difference). The phase contrast microscope observes living cells and unstained specimens by changing this phase difference and using the diffraction and interference phenomena of light to convert the phase difference into amplitude difference. Phase contrast is a light microscopy technique used to enhance the contrast of images of transparent and colourless specimens. It enables visualisation of cells and cell components that would be difficult to see using an ordinary light microscope.

As phase contrast microscopy does not require cells to be killed, fixed or stained, the technique enables living cells, usually in culture, to be visualised in their natural state. This means biological processes can be seen and recorded at high contrast and specimen detail can be observed. Fluorescence staining can be used in combination with phase contrast to further improve the visualisation of samples.

Phase contrast is ideal for thinner samples, therefore an inverted microscope system can be used. This provides the additional advantage of having more working space. Phase contrast can also be installed on upright microscopes.

If thicker samples need to be visualised in high-resolution, differential interference contrast (DIC) is a more suitable technique to use.

Components Needed for Phase Contrast

It is relatively simple and inexpensive to adapt an inverted or upright light microscope for phase contrast. The following components need to be installed:

● Phase contrast condenser with a phase annulus (also known as a phase ring)

● Phase contrast objectives - a set of objectives which each contain a phase plate.

● Centering Telescope Eyepiece 10x

There are 2 kinds of phase contrast condenser. The simple one has a socket for phase contrast slides, users need to exchange slides for different objectives during observation. It is a little bit complicated in operation, but users can choose 40x phase contrast objective & slide as a minimum set of phase contrast at a low cost, which is the most useful set in daily work.

The turret phase contrast combines all four phase rings inside. It makes switch rings according to objectives easier, but the full set phase contrast attachment cost is higher.

How to Use Phase Contrast Microscope

Step 1.

Turn the turntable phase contrast condenser turntable to the "○" mark, and adjust the instrument referring to the bright field observation method.

Put the phase contrast objective lens into the optical path, turn the phase contrast condenser turret, and turn the corresponding magnification scale to the front side. When the position is clearly felt, it means that the phase contrast ring plate has reached the desired position, and phase contrast observation can be performed

Take observation with a 10× phase-contrast objective lens as an example.

Put the 10× phase-contrast objective lens into the optical path, and push the corresponding hole of the insert plate to the position, or turn the turntable to the “10×” mark, and turn the aperture diaphragm to the maximum.

Remove the eyepiece on one side and replace it with the centering eyepiece, loosen the fixing screw, and twitch the upper part of the eyepiece, and observe the field of view at the same time, so that the image of the phase contrast plate and phase contrast ring plate is clear. Then tighten the set screw

Push the two adjustment handles into the adjustment hole to adjust the position, turn the two adjustment handles, adjust the position of the ring plate, and make the bright ring of the ring plate coincide with the dark ring of the phase plate.
